ABSTRACT

Esophageal squamous cell carcinoma (ESCC) is one of the leading causes of cancer death worldwide. It is urgent to develop new drugs to improve the prognosis of ESCC patients. Here, we found benzydamine, a locally acting non-steroidal anti-inflammatory drug, had potent cytotoxic effect on ESCC cells. Benzydamine could suppress ESCC proliferation in vivo and in vitro. In terms of mechanism, CDK2 was identified as a target of benzydamine by molecular docking, pull-down assay and in vitro kinase assay. Specifically, benzydamine inhibited the growth of ESCC cells by inhibiting CDK2 activity and affecting downstream phosphorylation of MCM2, c-Myc and Rb, resulting in cell cycle arrest. Our study illustrates that benzydamine inhibits the growth of ESCC cells by downregulating the CDK2 pathway.

ABSTRACT

The identification of tumor-related microRNAs(miRNAs)exhibits excellent promise for the early diag-nosis of cancer and other bioanalytical applications. Therefore,we developed a sensitive and efficient biosensor using polyadenine(polyA)-mediated fluorescent spherical nucleic acid(FSNA)for miRNA analysis based on strand displacement reactions on gold nanoparticle(AuNP)surfaces and electrokinetic signal amplification(ESA)on a microfluidic chip.In this FSNA,polyA-DNA biosensor was anchored on AuNP surfaces via intrinsic affinity between adenine and Au.The upright conformational polyA-DNA recognition block hybridized with 6-carboxyfluorescein-labeled reporter-DNA,resulting in fluores-cence quenching of FSNA probes induced by AuNP-based resonance energy transfer.Reporter DNA was replaced in the presence of target miRNA,leading to the recovery of reporter-DNA fluorescence.Sub-sequently,reporter-DNAs were accumulated and detected in the front of with Nafion membrane in the microchannel by ESA.Our method showed high selectivity and sensitivity with a limit of detection of 1.3 pM.This method could also be used to detect miRNA-21 in human serum and urine samples, with re-coveries of 104.0%-113.3%and 104.9%-108.0%,respectively.Furthermore,we constructed a chip with three parallel channels for the simultaneous detection of multiple tumor-related miRNAs(miRNA-21,miRNA-141,and miRNA-375),which increased the detection efficiency. Our universal method can be applied to other DNA/RNA analyses by altering recognition sequences.

ABSTRACT

Objective To evaluate the effect of intrathecal morphine and fentanyl on the level of natural killer(NK)cells in the hippocampus of rats with incisional pain. Methods Ninety-six healthy male Sprague-Dawley rats, weighing 180-220 g, aged 6-8 weeks, in which intrathecal catheters were successfully implanted, were divided into 4 groups(n=24 each)using a random number table: control group(C group), incisional pain group(P group), intrathecal morphine-fentanyl group(MF group) and intrathecal morphine-fentanyl plus incisional pain group(MFP group). A 1-cm longitudinal incision was made through skin, fascia and muscle of the plantar aspect of the right hindpaw in sevoflurane-anesthe-tized rats. At 20 min before establishment of the incisional pain model, the mixture of morphine 5 μg∕kg and fentanyl 025 μg∕kg was intrathecally injected in MFP group, while the equal volume of normal saline was given instead in NS and P groups. Six rats were selected at 24 h before establishment of the model (T0, baseline)and at 1, 6, 24, 48 and 72 h after establishment of the model(T1-5), and the mechani-cal paw withdrawal threshold(MWT)and thermal paw withdrawal latency(TWL)were measured. Six rats from each group were selected at T0, T2, T3and T5, venous blood samples were taken from the pe-ripheral vein, the animals were then sacrificed immediately, and the hippocampi and spleen were quickly harvested. The levels of NK cells in peripheral blood, hippocampi and spleen were measured. Results Compared with group C, the MWT was significantly decreased, the TWL was shortened, the levels of NK cells in hippocampi were increased at T2-5, the levels of NK cells in the spleen were increased at T2and de-creased at T3,5, and the levels of NK cells in peripheral blood were decreased at T2,3in group P(P<005), and the MWT was significantly decreased, the TWL was shortened, the levels of NK cells in hipp-ocampi were increased at T2,5(P<005), no significant change was found in levels of NK cells in hipp-ocampi at T3(P>005), the levels of NK cells in the spleen were increased at T2and decreased at T5, the levels of NK cells in peripheral blood were decreased at T2,5(P<005), and no significant change was found in levels of NK cells in the spleen and peripheral blood at T3in group MFP(P>005). Compared with group P, the MWT was significantly increased, the TWL was prolonged, the levels of NK cells in hippocampi were decreased at T2-5, and the levels of NK cells in the spleen and peripheral blood were de-creased at T2-5and increased at T3in group MFP(P<005). Conclusion Intrathecal morphine and fenta-nyl can maintain the level of NK cells stable in the hippocampus, which may be helpful for maintenance of immune function of rats with incisional pain.

ABSTRACT

Objective:To explore the value of MR-cine for assessment of the duodenum peristalsis in the patients with functional dyspepsia.Methods:25 patients with functional dyspepsia were selected according to the diagnostic criteria of RomeⅢ of functional gastrointestinal disorders (FGIDs)as case group and 25 cases of healthy volunteers who had been screened out in clinic were used as normal control group. After fasting for 8 h,supine position was performed.1.5TGEHDxMR was used to scan the coronal, axial and oblique coronal fast steady state precession (FIESTA)sequence of gastric and duodenal descending part. The images of 5 and 15 min after drinking 600 mL mannitol solution (concentration 2.5%)were collected and sent to MR AW4.4 workstation. The duodenal motility in the patients with functional dyspepsia was measured by recording the times of duodenum and measuring and calculating the percentage occlusion of duodenum contractions (PDC). Results:After drinking mannitol, the oblique coronal scan of all subj ects clearly displayed the anatomical structure and peristaltic wave of descending part of duodenum. After drinking 5 and 1 5 min of mannitol, the peristaltic wave frequencies in case group were lower than those in normal control group. The descending duodenum PDC of the patients in case group was significantly lower than that in normal control group (P0.05).Conclusion:MR-cine can evaluate preliminarly the duodenum peristalsis of the patients with functional dyspepsia. The diagnosis of FGIDs can be further studied by using the noninvasive MR-cine examination technique.

ABSTRACT

OBJECTIVE:To study the effects of propofol on invasion of hepatoma carcinoma HepG2 cell. METHODS:MTT method was used to detect the viability of HepG2 cells which were cultured with 0(negative control),1,3 and 10 μg/ml propofol for 48 h. The ability of cell invasion was detect by Tranaswell method. The phosphorylation level of nuclear factor-κB p65(NF-κB p65) and the expression of matrix metalloproteinase 2 (MMP-2),MMP-9,E-cadhherin and Snail were detected by Western blot method. RESULTS:Compared with negative control,1,3,10 μg/ml propofol inhibited the cell viability and invasion,down-regu-lated the expression of Snail and the phosphorylation level of NF-κB p65,and up-regulated the expression of E-cadherin (P<0.01). 3,10μg/ml propofol down-regulated the expression of MMP-2 and MMP-9(P<0.01). Above effects depended on drug con-centration(P<0.01). CONCLUSIONS:Propofol can suppress HepG2 invasion, which might be related to the inhibition of NF-κB/Snail signal pathway.

ABSTRACT

<p><b>OBJECTIVE</b>This study aims to observe the expression of Sclerostin during movement of orthodontic teeth and determine the effect of this protein on remodeling of periodontal tissues.</p><p><b>METHODS</b>Twenty-four Wistar rats were chosen. Orthodontic forces were applied between the bilateral incisor and first molar to achieve mesial movement. Rats in each group were executed at different time points (0, 1, 3, 5, 7, 14 d). Morphology of periodontal tissue was observed by hematoxylin-eosin (HE) staining. The number of osteoclasts were observed by tartrate-resistant acid phosphatase (TRAP) staining. Sclerostin expression were observed by immunohistochemical staining.</p><p><b>RESULTS</b>HE staining revealed that the resorption of alveolar bone intensified with prolonged movement. Results of immunohistochemical and TRAP staining revealed that Sclerostin expression and number of osteoclasts were related to duration of movement of orthodontic tooth. After staining for 5 days, the number of osteoclasts and Sclerostin expression reached their peak and then began to decline. The numbers of osteoclasts and the expression level of Sclerostin were higher at the compressive side than those at the tensive side.</p><p><b>CONCLUSION</b>Sclerostin affected orthodontic tooth movement by inhibiting the Wnt signaling pathway and by indirectly or directly controlling bone morphogenetic protein.</p>

ABSTRACT

Objective:We aimed to investigate the effect of neo-adjuvant chemotherapy on postoperative cognitive dysfunction (POCD) in elderly patients who underwent one-lung ventilation (OLV) surgery. Methods:A total of 90 esophageal carcinoma patients aged 60 years old or older were included. These patients were scheduled for esophagectomy, including two or three-field lymphadenec-tomy, and were randomly divided into two groups based on the American Society of Anesthesiologists status (Ⅰ or Ⅱ) and the Tu-mor-Node-Metastasis (TNM) classification stage (ⅡorⅢ), as follows:the neo-adjuvant chemotherapy group (Group N:n=45) that re-ceived preoperative neo-adjuvant chemotherapy;and the control group (Group C:n=45) that did not receive chemotherapy. The neuro-psychological test was performed 1 d before and 7 d after surgery to evaluate the changes in cognitive function. The incidence of POCD was also determined via the Z-value method in the two groups. Results:A total of 44 patients in Group N and 41 patients in Group C completed the neuropsychological tests. No statistical differences were observed in the demographics, TNM stage, and the intra-and post-operative clinical data between the groups. POCD was observed in 21 of the patients in Group N (47.7%) and 11 of the patients in Group C (26.8%), and the differences were significant (χ2=3.949, P=0.047). Conclusion:Neo-adjuvant chemotherapy can aggravate the impairment of cognitive function in the elderly patients undergoing OLV surgery and can significantly increase the incidence of POCD.

ABSTRACT

To investigate the effectiveness of the Shikani optical stylet [SOS] and Glide Scope video laryngoscope [GVL] for tracheal intubation of thyroid tumor patients with a difficult airway. One hundred and twenty thyroid tumor patients with a difficult airway, who were undergoing elective surgery requiring general anesthesia, were enrolled in the study. They were randomly allocated to 3 groups [n = 40 each] who underwent direct laryngoscopy [DL], SOS or GVL. The outcomes recorded were time to intubation, first-attempt success rate, mean artery pressure [MAP], heart rate [HR] and incidence of complications. The mean time to intubation in the SOS group [group S; 42.4 +/- 24.1 s] and the GLV group [group G; 29.8 +/- 22.3 s] was significantly less than that in the DL group [group D] [68.8 +/- 26.6 s]. The first-attempt success rate in group S [90.0%] and group G [97.5%] was significantly higher than that in group D [75.0%; all p < 0.05]. The HR and MAP at 1 min after intubation were lowest in group S [76.4 +/- 9.2 beats/min and 12.9 +/- 1.1 kPa], followed by group G [79.9 +/- 9.3 beats/min and 13.0 +/- 0.9 kPa] and then group D [90.4 +/- 8.1 beats/min and 16.6 +/- 1.2 kPa]. The difference was statistically significant [all p < 0.05]. The incidence of lip or mucosal trauma was lowest in group S, followed by group G and then group D. The SOS and the GLV had advantages over the DL in the management of thyroid tumor patients with a difficult airway in terms of a shorter time to intubation, a higher first-attempt success rate and a reduced incidence of complications. Thus, a rational choice of one of these techniques may be better for the perioperative safety of thyroid tumor patients with a difficult airway

ABSTRACT

Anthracycline-based antitumor drugs are one of the highly effective broad-spectrum anti-cancer drugs that are commonly used clinically. However, anthracyclines have the serious side effect of cumulative dose-related cardiotoxicity. Clinically, intake of myocardial protective drugs is one of the most important strategies for the safe use of anthracyclines in chemotherapy. Numerous cancer patients un-dergoing chemotherapy also require surgery, during which general anesthesia is commonly used. Various studies have shown that propofol and sevoflurane, the most commonly used general anesthetics, present a significant protective effect in myocardial ischemia-reperfusion damage. Therefore, for patients who have just undergone chemotherapy with anthracyclines, anthracycline-induced myocardial injury may occur at the acute or chronic phase of surgery. Consequently, if the effect of anesthetics on the cardiotoxicity of anthracyclines is similar to the protective effect of these drugs for myocardial ischemia-reperfusion damage, this protective effect on cardiotoxicity will have significant impact on the recovery of cardiopulmonary function of patients after surgery. Thus, further studies are needed to establish the effect of anesthetics on the cardiotoxicity of anthracyclines.
